Synthesis of Moracin C and Its Derivatives with a 2-arylbenzofuran Motif and Evaluation of Their PCSK9 Inhibitory Effects in HepG2 Cells

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a key factor in several cardiovascular diseases, as it is responsible for the elevation of circulating low-density lipoprotein cholesterol (LDL-C) levels in blood plasma by direct interaction with the LDL receptor. The development of orally available drugs to inhibit this PCSK9-LDLR interaction is a highly desirable objective. Here, we report the synthesis of naturally occurring moracin compounds and their derivatives with a 2-arylbenzofuran motif to inhibit PCSK9 expression. In addition, we discuss a short approach involving the three-step synthesis of moracin C and a divergent method to obtain various analogs from one starting material. Among the tested derivatives, compound 7 (97.1%) was identified as a more potent inhibitor of PCSK9 expression in HepG2 cells than berberine (60.9%). These results provide a better understanding of the structure–activity relationships of moracin derivatives for the inhibition of PCSK9 expression in human hepatocytes.     

以尿素为氮源合成AlN粉体过程中非晶碳的石墨化

针对有机合成过程中碳及碳化物的残余,传统方法中普遍使用除碳的工艺,而很少有文章针对非晶碳的结构和形貌进行表征。为此,本文采用高尿素含量的前驱盐体系,通过在氮气保护气氛中煅烧获得AlN粉体。采用X射线衍射分析、红外和拉曼光谱分析、扫描电子显微镜和透射电子显微镜对850~1 500 ℃温度范围内煅烧获得产物的结构和形貌进行表征,对AlN合成过程中含碳产物结构形貌的变化,以及AlN和含碳产物之间相互的依存生长关系进行分析。结果表明,AlN生长的过程中伴随着无定形碳的石墨化转变,AlN颗粒的形貌也受含碳残余产物形貌的影响而出现有规律的变化。     

Cyclopentadienyl rhodium(III) complexes as catalysts for the insertion of phenyldiazoacetate into E−H bonds

Rhodium(III) complexes catalyze the insertion of carbenoids generated from diazo compounds into E?H bonds (E = B, Si, N, but not C), although less efficiently than classical rhodium(II) carboxylates, despite formally higher oxidation state of the metal.     

Utility of 2-Diphenylphosphoryloxy-1,3-Dienes in Multicomponent Hetero-Diels–Alder Reactions to Construct Functionalized Six-Membered Nitrogen Heterocycles

The utility of 2-diphenylphosphoryloxy-1,3-dienes for the construction of substituted six-membered nitrogen heterocycles is presented. These dienes undergo boron trifluoride-promoted aza-Diels–Alder reactions when reacted with imines or related species formed in situ using aldehydes and amine derivatives. The stability of the dienes allows this three-component reaction to be carried out with no special precautions to eliminate water or air. Thirty-one examples of this process are presented. The usefulness of the enol phosphate functional group is highlighted in further reactions after the cycloaddition step to generate functionalized piperidenes or pyridines.     

Synthesis and in vitro antifungal activity of selenium-containing chitin derivatives

Mild and ‘green’ ultrasound-assisted reaction of chitin with 3-(chloromethyl)[1,2,4]selenadiazolo[4,5-a]pyridin-4-ium bromide in water affords novel selenium-containing cationic chitin derivatives. The thus obtained chitin derivatives are water soluble and are characterized by high in vitro antifungal activity comparable with conventional antifungal drug Amphotericin B.     

Effect of pressure on the low-temperature reaction of ethylene with N2O4

Calorimetric monitoring of the autoclave reaction N₂O₄ + C₂H₄ at –85 to +10 °C under argon pressure 10–30 bar revealed that the exothermic chemical reaction started at temperatures above –52 °C at 10 bar, whereas an intensive exothermic reaction started at –85 °C and pressure of 30 bar. IR study showed that oligo/polynitroethylene was formed at 30 bar, while carbonyl and hydroxy compound as well as nitrate R–ONO₂ formation occurred upon processing at 10 bar.     

A facile route to germanium(IV) binaphthoxide complexes: crystal structure of a chiral resolved germanium(IV) binaphthoxide

A method for the facile synthesis of chiral germanium(IV) binaphthoxide complexes from the corresponding binaphthols and an organogermanium trichloride has been developed, which allows these unusual types of compounds to be synthesized in high yields. The crystal structure of one such complex, (S)-[Ge{O2C20H10(SiMe3)2-3,3'}{Cl}{Ph}], has been determined.     

Synthesis of arsonium salts and betaines based on triphenylarsine and ω-bromoalkanoic acids

The reactions between Ph₃As and ω-bromoalkanoic acids with the length of polymethylene fragment n = 5, 7, 9 afford quaternary arsonium salts. Their treatment with alkali gives biologically active arsenobetaines. Solvent-free reactions of Ph₃P or Ph₃As with 2,3-dibromopropionic acid lead to the corresponding arsonium and phosphonium salts bearing (CH₂)₂COOH substituent.     

Photocatalytic reduction of fluoroalkyl-substituted alcohols activated by pentafluoropyridine

A new method for deoxygenation of fluoroalkyl-substituted alcohols involves derivatization of the hydroxy group with pentafluoropyridine followed by photoredox catalyzed reduction of the obtained hetaryl ethers using γ-terpinene as a source of hydrogen. The initial alcohols can be easily obtained by nucleophilic fluoroalkylation of the corresponding aldehydes.     

基于捕集柱阵列的制备型二维液相色谱纯化烟草中的4个组分

二维液相色谱(2D-LC)因具有较高的峰容量,在复杂样品的分离分析中获得了广泛的关注。然而,制备型2D-LC以纯化高纯单体为目标,在方法开发和设备构成等方面与分析型2D-LC有较大的不同,目前尚未得到充分的开发,在大规模的制备纯化中应用较少。本文以一套制备液相色谱模块为分离系统,以稀释泵、切换阀和捕集柱阵列为接口,构建了新型的制备型2D-LC系统,旨在规模化纯化多个活性成分。以烟叶中可以用作医药原料的烟碱、绿原酸、芦丁和茄尼醇等组分为目标物,考察了不同类型填料对样品的捕集效率、过载条件下的色谱保留行为等,优化了制备色谱条件。进而利用在线2D-LC系统实现了烟叶提取物的纯化,通过一次运行获得了4个高纯化合物。该系统具有中压色谱纯化成本低、系统在线运行自动化程度高、稳定性好及容易放大等优点。烟叶中活性化学成分的回收利用对促进烟草行业的发展及带动地方农业经济开发具有重大的意义。
